Fear learning triggers structural changes at GABAergic synapses in the basal amygdala

چکیده
Background In Pavlovian fear conditioning, in which initially a conditioned stimulus (CS) is repeatedly paired with an aversive stimulus (US), animals learn to associate CS with US, hence attaching emotional significance to sensory stimuli. Conditioned fear can be suppressed when the CS is repeatedly presented alone, a phenomenon known as fear extinction. Previous studies suggest that the mechanisms underlying fear conditioning involve inhibitory neurotransmission through GABAA receptors. GABAA receptors are generally composed of two a, two b and one g subunits. Sixteen GABAA receptor subunits have been identified in mammals, which indicates that these receptors may have multiple biophysical and pharmacological properties. Recent work showed that fear conditioning induces a dramatic downregulation of benzodiazepine binding sites and transcripts for gephyrin and some GABAA receptor subunits in the basal nucleus of the amygdala (BA), which were restored to control levels after fear extinction.

Fear learning induces structural and functional plasticity at GABAergic synapses in the basolateral amygdala
Background Previous work has suggested that alterations in GABAergic function within the amygdala underlie fear learning. In particular, it has been shown that Pavlovian fear conditioning induces a downregulation of benzodiazepine binding sites as well as transcripts for gephyrin and some GABAA receptor subunits in the basal nucleus of the amygdala (BA), which were restored to control levels af...
متن کاملASIC-dependent LTP at multiple glutamatergic synapses in amygdala network is required for fear memory
Genetic variants in the human ortholog of acid-sensing ion channel-1a subunit (ASIC1a) gene are associated with panic disorder and amygdala dysfunction. Both fear learning and activity-induced long-term potentiation (LTP) of cortico-basolateral amygdala (BLA) synapses are impaired in ASIC1a-null mice, suggesting a critical role of ASICs in fear memory formation. In this study, we found that ASI...
متن کاملStructural and functional characterization of dendritic arbors and GABAergic synaptic inputs on interneurons and principal cells in the rat basolateral amygdala.
The basolateral amygdala (BLA) is a complex brain region associated with processing emotional states, such as fear, anxiety, and stress. Some aspects of these emotional states are driven by the network activity of synaptic connections, derived from both local circuitry and projections to the BLA from other regions. Although the synaptic physiology and general morphological characteristics are k...
متن کاملInterplay of Amygdala and Cingulate Plasticity in Emotional Fear
The amygdala is known to be a critical brain region for emotional fear. It is believed that synaptic plasticity within the amygdala is the cellular basis of fear memory. Recent studies demonstrate that cortical areas such as the prefrontal cortex (PFC) and anterior cingulate cortex (ACC) may also contribute to the formation of fear memory, including trace fear memory and remote fear memory. At ...
متن کاملCompetition among model lateral amygdala principal cells during Pavlovian fear conditioning
Inputs about the conditioned (CS) and unconditioned (US) stimuli converge in the lateral amygdala (LA) during classical fear conditioning (FC). This leads to potentiation of CS inputs to LA neurons, and consequent increases in the firing of central medial amygdala (CeM) cells. In turn, CeM cells drive conditioned fear responses via their projections to fear effector neurons. However, transmissi...
